Ribosom čita jedan po jedan triplet, na svaki triplet se veže tRNA sa odgovarajućim antikodonom. tRNA otpušta aminoskiselinu koju nosi, koju ribosom zatim veže uz prijašnju aminokiselinu pomoću enzima peptidil transferaze koji katalizira stvaranje kovalentnih veza između dvaju aminokiselina, čime se tvori polipeptidni lanac.&amp;lt;ref name=&amp;quot;:1&amp;quot; /&amp;gt;&lt;br /&gt;
